Title: 應用基因重組大腸桿菌生產盤尼西林醯胺酵素

Abstract: 利用重組之大腸桿菌發酵可以生產出盤尼西林PGA,主要以大腸桿菌發酵液經過破細胞,上層液即為PGA酵素。本實驗中所使用隻菌種為基因重組大腸桿菌。其帶有 PGL-5 質體可以生產盤尼西林醯胺酵素,為了發酵得到高效率的PGA,利用菌體的生產總量與產物PGA成正比關係,探討各種培養基成分濃度與發酵條件下發酵培養對 PGA 酵素活性之影響,即了解各種培養基成分與培養條件對發酵的影響。由實驗結果得知,當Glucose碳源與Yeast氮源增加時,PGA活性之量也隨之增加;當轉速減少時PGA活性之量也減少;當培養溫度升高時,PGA量則降低。
